- Peter Parker is bitten by a radioactive spider and gains the abilites of a spider. When he fails to stop a burglar who runs past him his uncle his killed as a result. He therefore swears never to shirk his responsibility again.

Stan Lee's editorial explains: "We are omitting the word ADULT from our masthead. A number of out teen-age readers have written to say that it makes them feel a bit awkward to buy a magazine which seems to be written exclusively for older readers."
Lee also explained the new feature: "..The SPIDERMAN, who will appear every month in AMAZING. Perhaps, if your letters request it, we will make his stories even longer, or have TWO Spiderman stories per issue."
Based on job numbers researched by Will Murray, the stories that appeared in Amazing Spider-Man #'s 1-2 were likely prepared for the next issues of Amazing Fantasy. Publisher Martin Goodman cancelled the title before sales figures came in and immediately reinstated Spider-Man in a title of his own when he discovered how well the issue sold.

- Dr. Doom tries to trick Spider-Man into helping him defeat the Fantastic Four. When Spider-Man turns him down, he decides to capture Spider-Man. Flash Thompson dresses up as Spider-Man for a prank and Dr. Doom mistakes him for the real Spider-Man and captures him instead, and Flash has to be saved by the real Spider-Man.

- Peter has the 24 hour flu when Doc Ock captures Betty Brant in hopes of luring Spider-Man into battle. Ock beats the under the weather web-head and unmasks him, but everyone thinks it is just Peter trying to be brave by impersonating Spidey. Once he has recovered, Spider-Man battles through the wild animals Ock has released from the zoo to take on old six-arms himself.

- Synopsis
- When the Circus of Crime comes to town, they advertise that Spider-Man will be appearing in an attempt to fill the big top before they rob it. Spider-Man sees the advertisements and decides he will show up. He is hypnotized along with the rest of the audience, but unfortunately for the Circus of Crime, the Daredevil is in attendance and immune to the Ringmaster's hat.

- Synopsis
- Spider-Man attends a fan club meeting organized by Flash Thompson, but the Green Goblin crashes the meeting. Fortunately the Human Torch is in attendance to help Spider-Man. During the battle Spider-Man finds out that Aunt May is in the hospital and he leaves, causing everyone to believe that he is a coward.
